Half a dozen of 1, 6 of another. Doesn’t matter as long as the enclosure is large enough for 2 subs and is air tight.

There are a few reasons to potentially have separate chambers, primarily if you will be driving each driver with a separate amp or amp channel. If not wired correctly or if 2 amp channels aren’t in sync, two drivers sharing the same chamber could fight each other and cause issues, potentially damage. But that is rare so I wouldn’t worry about it. If you can, use a single amp channel to drive both subs, just make sure you wire them up correctly.
