If I have a two channel amp that I have designed for two subs (ohms,rms, etc) would it damage the amp or sub to only hookup one channel for a while if I only have one sub?

Nope, would do no harm at all.

If you have a 2 ohm stable stereo amp and wire a dual 2 ohm sub to 1 ohm and attach it to the amp stereo things might not work out too well...

model numbers of said items might help

I understand all that. It is a dual 4 ohm sub (wired at 2ohms) attached to a 2ohm amp (2 ohms per channel).

Sub is Jl w6v2 10
Amp to be determined
