Running Dynaudio 342 system 2 way active (6.5 mid bass + Tweeter) right now and hope to add the 3" mid dome in the future.

The 3" mid dome come set in a moulded hard backing enclosure similar to a tweeters cup. With this design I wondered how much it matters what the enclosure behind it is. Could it just be flush mounted to a sail panel or flat kick panel with no additional effort to seal it off?

There is a built in enclosure on the md142/2. It is integrally sealed like most tweeters. The driver sees nothing outside of its own enclosure, so as long as it is mounted securely, you are good to go.

Yep. No enclosure needed. It's actually a very nice mid for a dome. I really liked mine. A little on the bright side for a Dyn but not really that bright. It's a very nice addition to that set. It likes a pretty high high pass, I ran mine at 950hz to 6khz. Sounded great like that.
